So what the heck is a peptide? A peptide is simply a compound made up of amino acids (2 or more). Basically, peptides are just tiny proteins. Your body is full of them, and they carry out many functions that help you survive. In the last few decades, scientists have been researching peptides in the hope of developing them into remedies to treat various illnesses. Melanotan 2 was first created as a possible preventative measure against skin cancer.

So now, let’s separate fact from fiction and take an unbiased, levelheaded look at this peptide.
Credit for the discovery of Melanotan 2 goes to a team of scientists headed by Drs. Victor J. Hruby and Mac E. Hadley at the University of Arizona. They were working on developing a drug aimed at preventing skin cancer using the body’s natural processes.
The body’s natural defense against sun damage? A tan. So the team sought to develop something that would stimulate the body’s own tanning response BEFORE exposure to sunlight and potentially harmful UV rays. This way the skin would be less likely to burn in the sun and thus less likely to develop cancer.
Quick tanning lesson: there are 2 different ultraviolet rays in sunlight – UVA and UVB. UVB rays burn the skin, and UVA rays cause tanning. How? UVA rays penetrate to lower levels of the skin, where they stimulate cells called melanocytes to produce melanin, that wonderful brown pigment that changes your skin tone from albino to bronze. In your body, a hormone called Alpha MSH (α-MSH) stimulates melanin production in the skin, a process called melanogenesis.
So the researchers figured that administering extra amounts of Alpha MSH to a patient would make them tan, even before sunlight exposure. And they were right…sort of. While it did work, regular Alpha MSH had way too short of a half-life to be effective as a drug. As a result, they needed to develop something with the same properties as Alpha MSH, but with a steadier, longer-lasting effect. That’s when they created a new peptide they dubbed “Melanotan.” Further refining of Melanotan led to the development of Melanotan 2, a peptide longer lasting and hundreds of times more potent that Alpha MSH, and one that held great promise as a future pharmaceutical.
We’ve established that Melanotan 2’s effects are legitimate and are backed up by numerous studies. However, are there any risks?
First thing’s first: Melanotan is usually well tolerated. It’s not the equivalent of playing Russian roulette with your body, like some mainstream news outlets would make it seem. But some side effects can occur if too large of a dose is administered. Therefore, it's important to start with a small dose and work up from there.
In clinical studies, any side effects reported were generally mild and short-lived. Importantly, any harsher side effects that did occur were correlated with extreme dosages, sometimes 6 or more times over the recommended maximum.
Now let’s take a clearheaded look at Melanotan 2’s side effects… Generally, the most common are tiredness, mild nausea, and facial flushing. These are often mild and usually go away shortly after injecting. In many studies, these were the only side effects observed.
While there have been more extreme cases of side effects outside of clinical studies, again, these were related to overdose. Crucially, it’s important to note that side effects are correlated with extremely high dosages taken at once, far beyond recommended protocols (more on this later). Still, this shows that overdose can have negative consequences, and should be avoided by using proper dosing protocols.
Study dosages for Melanotan 2 have typically ranged from 0.01 milligrams per kilogram (mg/kg) of body weight to 0.03 mg/kg. In the pilot phase-1 clinical study mentioned before, the research team recommended a single dose of 0.025 mg/kg every day for future Phase 1 studies. So, for an 80 kilogram person, a study dose would call for about 2 milligrams, given once daily. A more conservative dosage (0.01 mg/kg) would be about 0.8 mg.
However, these recommendations are quite high; maximum recommended dosage for a single day is 1 milligram, and this amount should be worked up to slowly, especially for new users. Importantly, many may not even need to use this much and will see great results from smaller daily dosages.
A much more effective method of administration is to deliver smaller dosages split up into separate loading and maintenance phases. For example, a month-long loading phase (4-6 weeks) would require 200 to 500 micrograms (mcg; 1mg = 1000mcg) given 3 to 5 times per week. During this phase, it is often recommended to start with a smaller dose at first to gauge tolerance to side effects, and then to increase slowly if required.

In peptide form, Melanotan 2 is a freeze-dried powder. To make it injectable, it has to be mixed with a sterile fluid (usually bacteriostatic water). This process is called “reconstitution.” So how is Melanotan 2 reconstituted?
First thing’s first: materials. A vial of bacteriostatic water, an insulin syringe, an alcohol swab, and a vial of Melanotan 2 (obviously) are needed. To start, both vial caps are removed, and each rubber stopper is wiped with alcohol. Then, the syringe is used to withdraw the bacteriostatic water. Generally, 1 mL of water is used per every 10 mg of Melanotan 2. Next, the bacteriostatic water is injected into the vial of Melanotan. This is done very slowly, though, and the water is aimed at the side of the vial instead of directly onto the dry powder. This prevents damaging the fragile peptide bonds. Finally, the vial is gently swirled to completely dissolve the peptide. Swirling rather than shaking the vial also prevents peptide bond damage.
An important point here is that it is used IMMEDIATELY after it’s been reconstituted. Freeze-dried (aka lyophilized) peptides can keep for weeks if they're kept refrigerated and can last months or years under the right storage conditions (i.e. frozen at -20 degrees Celsius or lower). But once reconstituted, it begins quickly losing potency. Even kept refrigerated, it will only stay usable for around 10-14 days. So it must be used ASAP.
